Obesity and disease risk
Population studies examined associations of obesity measures and metabolic health with cancer and mortality, COVID-19 severity and mortality, progression from prediabetes to diabetes, and outcomes among patients undergoing cardiac resynchronization therapy. In the Kailuan cohort, BMI, waist circumference, and metabolic health were combined into six obesity phenotypes and analyzed with Cox models for cancer risk and all-cause mortality. In Japanese hospitalized patients with COVID-19, critical illness increased monotonically with BMI, while mortality followed a J-shaped association. Among people with HIV aged 40 years or older who had prediabetes, higher BMI and waist circumference were associated with increased risk of progression to diabetes. Among patients undergoing cardiac resynchronization therapy, BMI-defined obesity was examined in relation to all-cause mortality, heart transplantation, left ventricular assist device implantation, periprocedural complications, and echocardiographic response (PMIDs 39479267, 38636933, 38923420, 39031161).

Drug-based weight management
These studies evaluated GLP-1βpathway drugs in populations including type 1 diabetes, type 2 diabetes, and obesity. In adults with type 1 diabetes and a mean BMI of 36.7 kg/mΒ², observational tirzepatide treatment was associated with HbA1c reductions of 0.45% at 3 months and 0.59% at 8 months. A phase 1 program evaluated the safety, pharmacokinetics, and pharmacodynamics of oral GLP-1 receptor agonist lotiglipron in participants with type 2 diabetes and/or obesity, with nausea the most frequently reported adverse event. A randomized trial of exenatide in overweight or obese adults treated with olanzapine found mean weight changes of β0.5 kg with exenatide versus +2.6 kg with placebo over 16 weeks (PMIDs 38317405, 38751362, 40203970).

Bariatric surgery outcomes
Clinical studies evaluated perioperative management, behavioral response, comorbidity outcomes, and long-term procedure performance in people with obesity. A 206-case study tested a systematic preoperative bariatric-surgery protocol for consultation efficiency, hospital stay, surgical duration, and costs; a prospective sleeve-gastrectomy cohort used control-of-eating questionnaires at baseline and 3, 6, and 12 months to compare patients achieving at least 25% total weight loss with those who did not. Other cohorts examined symptom and intracranial-pressure outcomes after bariatric surgery in idiopathic intracranial hypertension and 10-year weight-loss, remission, and complication outcomes after biliopancreatic diversion with duodenal switch or single-anastomosis duodeno-ileal bypass with sleeve gastrectomy (PMIDs 39444955, 38502520, 39365468, 40962919).

Adipose and gut mechanisms
Preclinical studies examined a yellow leaf green tea extract, a hyaluronic-acid microneedle delivery system, and signaling mechanisms related to adiposity, inflammation, lipid synthesis, and thermogenesis. Yellow leaf green tea extract reduced body weight, hepatic inflammation, fat accumulation, and hyperlipidemia in high-fat-diet C57BL/6J mice while activating AMPK/CPT-1Ξ± and inhibiting FAS/ACC/SREBP-1c; fecal microbiota transplantation was used to investigate the contribution of altered gut microbiota. A fast-soluble hyaluronic-acid microneedle matrix containing a weak acid-degradable oleanolic acid dimer of rosiglitazone released rosiglitazone in obese mice; oleanolic acid improved the inflammatory status of obese adipose tissue, and the treatment promoted white-adipocyte browning. Hyperforin promoted adipose thermogenesis through a DlatβTrpv3βAMPK pathway, whereas the MIF tautomerase inhibitor 4-IPP promoted mitochondrial thermogenesis and adipocyte browning through mechanisms involving CD137 and PI3K/AKT signaling (PMIDs 38407390, 38941683, 39631519, 41741765).

Predicting obesity treatment response
Several studies evaluated biological or functional markers related to monogenic obesity diagnosis or weight-loss outcomes. Monogenic-obesity testing in children and adolescents reassessed variants in genes including LEPR, POMC, MC4R, and PCSK1 using updated software interpretation and literature review, improving classification certainty. A pilot sleeve-gastrectomy cohort analyzed gut microbiome and genetic profiles in relation to 12-month excess-weight-loss success and identified an association between the genus Akkermansia and weight loss, while another study evaluated preoperative six-minute-walk-test measures as predictors of early nonresponse after metabolic and bariatric surgery (PMIDs 39462520, 40533342, 41657084).
